Microarray Assay - Biochip - Development Scientists - Raleigh, NC

Description
Join team developing protein microarrays, which are miniaturized and parallel assay systems that contain small amounts of purified proteins in a high-density format which provide simultaneous determination from a diversity of analytes using small samples within a single test. We seek an individual to contribute to the development of such "biochips" which include a substrate having a surface spotted with virus-like particles or capsid fragments bound to discrete locations on its surface.
When illuminated with polarized light at a wavelength and angle appropriately tuned to the chip, perturbation of near-null reflectance condition due to target binding is readily observed as an increase in the reflected intensity allowing for parallel detection of analytes with high sensitivity and dynamic range and consequently detection of proteins in human serum. Most importantly, no labeled detection reagents are needed beacuase the analyte is detected directly.
Direct detection of pathogen-derived analytes, proteins or nucleic acids can provide for effective diagnostics, especially when multiplexing many analytes with a rapid detection method that is inexpensive.
System Components:
*** A substrate surface with proteins, nucleotides, virus-like particles or capsid fragments, bound to discrete locations on the surface of the substrate, that present conformational epitopes (biochip).
*** A light source that illuminates the biochip.
*** A reflected light detector that measures binding-induced changes on the surface of the biochip.
Seeking individuals who are familiar with TWO or more of the following:
*** Label-free detection techniques
*** Spectroscopic ellipsometry and surface plasmon resonance.
*** Splicing regulator proteins & pathogenic RNA targets
*** Depositing reproducible layers onto silicon dioxide surfaces.
*** Piexoelectric protein printing.
*** Investigating additives to improve spot morphology in protein arrays.
*** Reflectance calculations.
*** Binding kinetics of protein-RNA interactions.
